Pathway
NF-κB signalling
Central transcriptional inflammatory pathway downstream of many innate receptors.
Also called: NF-κB, nuclear factor kappa B

In short
- 1What it is: a pathway in signalling and mechanisms.
- 2How it connects: acts on 0 and is affected by 9 concepts — mostly 1 molecule, 5 receptor, 2 pathway.
- 3Where it sits: signalling on the 15-step immune chain.
What it does
Downstream of TLRs (MyD88→IRAK4→TRAF6→IKK→IκB phosphorylation→NF-κB nuclear), TNFR1, IL-1R, BCR, TCR; drives TNF, IL-6, IL-1β, IL-8, IL-12, COX-2, VCAM-1, E-selectin gene transcription; suppressed by glucocorticoids (broad NF-κB suppression = mechanism of steroid anti-inflammatory effect)
