Therapy
Dupilumab
Monoclonal antibody blocking IL-4/IL-13 receptor signalling.
Also called: dupilumab, Dupixent, DUPI

In short
- 1What it is: a therapy in therapies and applications.
- 2How it connects: acts on 3 and is affected by 4 concepts — mostly 1 receptor, 2 disease, 2 trial.
- 3Where it sits: therapy on the 15-step immune chain.
What it does
Human IgG4κ; anti-IL-4Rα → blocks both IL-4 (Th2 differentiation, IgE class switching) and IL-13 (goblet cells, smooth muscle, FLG suppression) simultaneously via shared Type II receptor subunit; broadest indication of any biologic: AD, severe asthma, CRSwNP, EoE, prurigo nodularis, COPD T2 (FDA 2024); Side effect: conjunctivitis 10-28% in AD (IL-4/13 regulate ocular surface goblet cells → mechanistic prediction, lubricating drops + ophthalmology)
