Molecule
Adenosine
Produced from ATP by CD39 (NTPDase1, extracellular) → AMP → CD73 (ecto-5'-nucleotidase) → adenosine; binds A2AR on T cells, NK cells, DCs → cAMP → suppression of effector function.
Also called: adenosine, A2AR ligand

In short

- 2How it connects: acts on 1 and is affected by 2 concepts — mostly 3 cell.
- 3Where it sits: failure, target on the 15-step immune chain.
What it does
Adenosine in TME: pathway includes Tregs (CD39+CD73) + tumour CD73; A2AR antagonists (AZD4635, ciforadenant) in Phase 1/2 ICI combinations; CD73 blockade (oleclumab, anti-CD73 Phase 2)
